There are several major and minor queer characters in Guild Wars 2, including a main character who is a queer woman, prominent lesbian characters, and one trans character. Guild Wars 2 was also highlighted by several respondents to the Queer Representation (2016) survey for its thorough character creation system, despite the lack of nonbinary gender options.

Caithe was the original lesbian protagonist in the game, with a girlfriend named Foalin. Foalin is a villain, and their relationship is emotionally abusive; other characters care about Caithe and the way she is treated, but don't have any issues with her attraction. This relationship and its plot arc was concluded after the Heart of Thorn expansion, but Caithe is still in the story.

Majory and Kasmeer are a lesbian couple, who have become central leads in the game since the 'Living World' update.

The Sylvari are considered pansexual, as they don't see gender in their relationships.

There are many gay NPCs within the game, mostly within sidequests.

The game also features many nonbinary / non-conforming characters, or instances of cross-dressing. The Charr and the Asura have options for women to wear traditionally masculine outfits, and are able to mix-and-match some masculine and feminine hair and faces. The Sylvari can also be made to look androgynous.
